Is this suitable for a ford transit long wheel base minibus?

I inflate the giant tractor tyres with mine, so most certainly - yes.

if I power the unit from the car battery, does this unit reach all 4 tyres with the cables supplied (MPV sized car)

The length of the lead and battery clips would give you just over 10 ft from the battery.There is also a 3 metre air extension air hose in the bag as well as the normal air hose which is 1ft 6inches. You would not have enough to walk round the back of the vehicle without using the air extension hose but otherwise should be OK.

Any reason why I couldn't connect main supply to 12v DC converter to operate from the garage?

I believe the 12 V DC Converter is designed to run off the 12V car circuit and not a 240V alternating supply. Suggest you ring customer service.
