What is the on/ off knob for?

It is a vent. If the case creates vacuum, with change of barometric pressure or temperature, it allows you to release the vacuum and open it.

will a couple of boxes of ammo fit inside with a gun?

Yes and no. If you care to modify the plastic carton the shells are in, you could probably squeeze 50+ in with your gun. It comes with a solid block of black foam to use as you like. However, if you're flying, they have to be contained, preferably in the original box. I took the foam out and stowed ammo, knife, and cleaning tools and took my unloaded gun in its original locking case. Using TSA locks on them in my checked bag, I went round trip without a hitch.

Will it fit an iwi masada?

Lock open your IWI 9mm and measure it’s total length. Verify the inside dimensions of the hard shell case. If your IWI is not longer than the inside dimensions of the hard shell case, then you can conclude that your handgun will fit. You make that decision based on measurements...not my response. I don’t carry a IWI. I carry a .380 Glock

does this require 1 lock or 2 locks?

You really only need one. The case is designed for 2 just in case.
